WHAT WE OFFER

We measure whole-plant gas exchange (photosynthesis, transpiration and stomatal conductance) in well-controlled conditions and its response to changes in abiotic factors and to exogenous application of the plant hormone abscisic acid (ABA).

 

We can rapidly modify the following factors and conditions (see below for specific ranges)  in the measurement cuvettes:

 

  • CO2 concentration: responses to CO2 enrichment and withdrawal

  • Relative air humidity (i.e. VPD)

  • Ozone concentration

  • Changes in white light intensity or application of darkness,  red and/or blue light.

  • Spray-application of ABA​

Different factors can also be applied sequentially or simultaneously.

 

Prior to gas-exchange measurements, we grow plants in approved growth chambers at standard conditions. Arabidopsis thaliana is routinely measured at the vegetative stage (plant age 23-30 days), and cereals at plant age of 10-20 days, but analyses of flag leaves are also possible.

 

Standard growth conditions for Arabidopsis cover 12/12 photoperiod, 23/18˚C temperature, 150 µmol m-2 s-1 light and 70% relative humidity. In case of cereals, growth light is increased to 500 µmol m-2 s-1.

 

Soil is 4:3 (v:v) peat: vermiculite mixture, its moisture is kept at 80% of maximum water capacity unless drought experiment is performed.

Specific Ranges

 

In experiments with Arabidopsis thaliana, we can change the conditions in the measuring cuvettes as follows:

  • CO2 concentration: 5 to 1300 ppm

  • Relative air humidity (i.e. VPD): approx 30-80%

  • Ozone concentration: 40-1000 ppb

  • White light intensity: 0-700 μmol m-2 s-1

  • Red light intensity: 0-1200 μmol m-2 s-1

  • Blue light intensity: 0-1800 μmol m-2 s-1

 

In experiments with other species, the ranges are:

  • CO2 concentration: 5 to 1300 ppm

  • Relative air humidity (i.e. VPD): approx 30-80%

  • Ozone concentration: 50-1500 ppb

  • White light intensity: 0-500 μmol m-2 s-1
